The secondary antibody is immobilized on the quality control line (line C) of the membrane. When there is a certain amount of FAM-biotin (or FITC-biotin) double-labeled probe in the sample, it forms a polymer of colloidal gold-FITC monoclonal antibody-FAM-biotin with colloidal gold. Capture, so that the T line develops color, and the C line captures the colloidal gold labeled with FITC monoclonal antibody and develops color; when the target molecule exists, the Cas protein is activated and non-specific ssDNA cleavage occurs, the dual-labeled probe is degraded, and the colloidal gold-FITC If the polymer of mAb-FAM cannot be captured by the T line, the T line will not develop color, and the C line can still capture the colloidal gold labeled with FITC mAb and develop color.
