      Cas14a1 nuclease is an endonuclease that specifically binds and cleaves target ssDNA under the guidance of tracrRNA:crRNA (or sgRNA) without the need for a PAM site. Compared with other Cas proteins, the molecular weight of Cas14 protein is generally smaller (400-700aa); similar to Cas12, Cas14a1 can also bind target nucleic acid and activate its ssDNA trans-cleavage activity, so it can be used for molecular detection of target nucleic acid; Unlike Cas12, Cas14a1 can only bind to ssDNA targets, so the amplified and enriched target nucleic acid needs to be treated with T7 exonuclease, and one of the amplification primers needs to be modified with phosphorothioate to ensure T7 exonuclease Dicer cleaves only one of the strands, leaving the ssDNA target strand for Cas14a1-mediated molecular detection.
